Pathos announces cancer drug partnerships with AstraZeneca and Jiangsu Alphamab

Pathos AI announced on Aug. 4 two new partnerships with AstraZeneca and Suzhou-based Jiangsu Alphamab Biopharmaceuticals, aimed at expanding the New York company's clinical cancer pipeline.

Under the agreement with Jiangsu Alphamab, a subsidiary of Alphamab Oncology, Pathos will pay $125 million upfront and could pay up to $2.093 billion in milestone payments for rights to a bispecific antibody-drug conjugate in late-stage development for breast cancer. The Chinese company is also eligible for tiered royalties ranging from high-single-digit to low-double-digit percentages based on annual net sales if the product reaches the market.

The financial terms of the partnership with AstraZeneca were not disclosed. This deal focuses on a preclinical PROTAC degrader targeting breast cancer. According to Pathos, both agreements will utilize its proprietary AI platform Foundry, which is described as being built on "the largest oncology foundation model in the industry." The company said Foundry uses thousands of AI agents to analyze complex clinical, biological, and real-world data to optimize trial design and dosing, as well as match patients to treatments most likely to provide therapeutic benefit.

For the AstraZeneca partnership, Pathos will use Foundry to advance AZD4241—a drug candidate that targets and degrades estrogen receptors—for ER+/HER2- breast cancer. Pathos will lead early-stage development of AZD4241 using insights from its AI platform; it remains unclear when AstraZeneca might assume further responsibility for development.

In its deal with Jiangsu Alphamab, Pathos has licensed JSKN016—a molecule identified through Foundry—which is currently undergoing pivotal development for later-line triple-negative breast cancer in China. Jiangsu Alphamab retains full rights over JSKN016 in China, Hong Kong, Macau, and Taiwan, but grants Pathos global rights elsewhere along with responsibility for worldwide development and commercialization informed by Foundry insights. "Every major asset-level decision at Pathos flows through Foundry," the company said.

With these agreements, Pathos now has four assets in clinical development: JSKN016; AZD4241; pocenbrodib (for prostate cancer, breast cancer, and multiple myeloma); and Phase 2 P-500 (for solid tumors).
