Review examines effects of amino acid restriction on aging and metabolism

A recent review published in Cell Press Blue suggests that restricting specific amino acids may reproduce some of the metabolic benefits of protein restriction, with potential implications for health and longevity, according to an article released on Aug. 3.

The review maps how certain amino acids shape metabolism and cellular aging but notes that laboratory findings are not yet ready to guide dietary advice. The authors highlight that while increasing protein consumption is widely promoted, especially for older adults, animal studies have shown that limiting intake of amino acids such as isoleucine, methionine, and valine can provide many benefits associated with protein restriction. These findings raise questions about current recommendations but do not establish that reducing specific amino acids will extend healthspan or lifespan in humans.

The review discusses evidence from human studies, laboratory animals, cells, and model organisms like yeast and fruit flies. It states, "In short-term studies, individuals consuming high-protein diets experienced greater weight loss than their peers... When combined with adequate protein intake, resistance exercise may help older adults preserve muscle function and reduce age-related frailty." However, it also points out associations between high protein intake and increased risks of diabetes, cancer, overall mortality, and cardiovascular mortality based on observational data such as the National Health and Nutrition Examination Survey (NHANES), though causation has not been established.

Short-term randomized controlled trials are cited as showing low-protein diets can lower blood glucose levels and body weight without calorie restriction. Limiting certain amino acids has improved insulin responsiveness in brief human studies; a four-week branched-chain amino acid (BCAA) restriction trial improved glucose homeostasis in people with metabolic syndrome. In animal models, dietary protein restriction reduced adiposity and improved longevity by increasing energy expenditure while improving glucose regulation—effects partly linked to increased fibroblast growth factor 21 (FGF21) expression after restricting various dietary proteins or BCAAs.

Other experimental evidence connects FGF21 to enhanced insulin sensitivity and reduced inflammation in tissues; mouse models have shown FGF21 overexpression extends lifespan. Restricting BCAAs for several weeks in humans led to better insulin sensitivity along with higher circulating FGF21 levels; cognitive benefits have so far only been observed in rodents. The review also describes how experimental restrictions or depletions of other amino acids impact key cellular signaling pathways but says dietary benefits for humans remain unclear.

The authors caution against applying these findings broadly: "Protein or amino acid restriction could be harmful for pregnant women, growing children...and older adults who already have inadequate protein intake." They note that much evidence comes from laboratory models rather than long-term human trials. Future research could clarify safe levels of individual amino acid restrictions while considering factors like age or health status.
